چکیده

Investigation of DNA damages induced by heavy ions is of great interest for a better understanding of the processes which appear in clinical radiotherapy and are responsible for the destruction of the cancer cells. The most serious damages of DNA molecules are doublestrand breaks (DSB), i.e. the rupture of both DNA strands in the range of a few base pairs and single-strand breaks (SSB), when one of the DNA strands is broken [1]. In particular, DSBs lower effectively the cell repair rate and leads often to cell death. Thus, heavy ion irradiation of tumour cells has been used very successful in cancer therapy. However, a detailed understanding of the processes which occur on the molecular level, leading to this effective cell death, is still lacking. Conventional methods, e.g. gel-electrophoresis measurements, are suitable to analyse the percentage of supercoiled and relaxed molecules and also linear DNA fragments, but it can not provide detailed information about the fragment distribution [2]. The latter can be achieved by scanning force microscopy (SFM) which permits direct visualisation and measurement of individual DNA fragments with an accuracy of a few nanometers. In this contribution we present a SFM analysis of radiation-induced damages in ФX174 plasmids in 20 mM Hepes solution after X-ray and C ion irradiation at a dose of 500 Gy. For sample preparation, 3μl plasmid solution in 10 mM MgCl2 [3] has been deposited on mica substrates and rested in air for 3 minutes. Subsequently, the substrates were washed with 400 μl double distilled water and dried with nitrogen. Nine samples have been prepared for each irradiated solution and around 10 images were recorded for each sample. The samples have been imaged in air with tapping mode using a CP-R microscope from Veeco Instruments. All images were recorded with a scan area of 3 μm x 3 μm and a resolution of 512 pixels x 512 pixels. The length of the fragments has been analysed using ImageJ software.

Induction of cancer stem-like cells in A549 cells after exposure to carbon ions and X-rays

Background: Cancer stem-like cells (CSCs) play a crucial role in the initiation, progression, and recurrence of cancer. Evidence indicates that the high linear energy transfer (LET) carbon ion beam is more effective against CSCs than the conventional X-ray beam. Carbon ion radiotherapy is considered as a promising cancer strategy, however, information about whether, or not, new CSCs are induced...

Effects of carbon ion irradiation and X-ray irradiation on the ubiquitylated protein accumulation

C-ion radiotherapy is associated with improved local control and survival in several types of tumors. Although C-ion irradiation is widely reported to effectively induce DNA damage in tumor cells, the effects of irradiation on proteins, such as protein stability or degradation in response to radiation stress, remain unknown. We aimed to compare the effects of C-ion and X-ray irradiation focusin...

Dramatic Increase in Oxidative Stress in Carbon-Irradiated Normal Human Skin Fibroblasts

Skin complications were recently reported after carbon-ion (C-ion) radiation therapy. Oxidative stress is considered an important pathway in the appearance of late skin reactions. We evaluated oxidative stress in normal human skin fibroblasts after carbon-ion vs. X-ray irradiation. Survival curves and radiobiological parameters were calculated. DNA damage was quantified, as were lipid peroxidat...

Sperm DNA damage in mice irradiated with various doses of X-rays alone or in combination with actinomycin D or bleomycin sulfate: an in vivo study

Background: DNA damage in male germ cells due to exposure to environmental and manmade physico-chemical genotoxic agents is considered as the main cause of male infertility. The aim of this study was to evaluate the effects of combined modalities (radiotherapy and chemotherapy) routinely used for cancer treatment on mouse sperm chromatin in vivo. Materials and Methods: Forty-eight mice were div...

Crystal Structure of Nonaaquayttrium(III) Bromate at 100 K

The structure of the nonaaquayttrium (III) bromate, [Y(H2O)9](BrO3)3, at low temperature (100 K) has been studied by means of single-crystal X-ray diffraction. Crystallography shows a hexagonal unit cell, space group P63/mmc (No. 194) with Z = 2, a = b = 11.7104(11) Å, c = 6.6259(5) Å and V = 786.90(12) Å3 at 100 K. The hydra...
